What can a diagnostic tool tell you?

A manufacturer interface such as the Shimano SM-PCE02 connects the bike to software on a computer and gives you the error history, the firmware version of each component and, for electronic groupsets, the shift settings. A multimeter tells you something more basic but often more useful: whether 36V is arriving where it should, whether a speed sensor lead has gone open circuit, and whether one cell group in a pack is dragging the rest down. Between them they separate a wiring fault from a component fault, which is the whole point.

Do I need a manufacturer interface, or will a multimeter do?

For dead-simple faults — a chafed wire, a corroded connector, a battery that will not take charge — a multimeter and continuity beeper find the problem for a fraction of the outlay. An interface is what you need for anything the system reports internally: a code that clears and comes back, a component that needs its firmware brought in line with the rest, or settings that only exist in software. Start with the meter, add the interface if the bike keeps telling you something you cannot read.

What should I check on a used interface or meter?

Inspect the connector on an interface cable pin by pin — the small plugs used for electronic groupset wiring bend easily, and one splayed pin makes the connection intermittent rather than dead, which is the hardest kind of fault to chase. Flex the cable gently at the moulded boot and watch for stiffness or a kink. Ask what the seller connected it to and on which operating system, since older interfaces can be left behind by driver changes. On a multimeter, test both ranges: a blown high-current fuse is common, leads crack at the plug end, and a meter that has been used on mains work may have had a rougher life than a bike ever gave it.

What is a fair price for used diagnostic kit?

Value here is binary — a cable that connects is worth having, a cable that does not is worth nothing — so favour sellers who can describe a session they ran with it rather than ones listing it as untested. Manufacturer interfaces hold their price better than general electronics because there is no alternative for the job. A basic multimeter is cheap enough new that used only makes sense if the leads and fuses are known good.
